Construction and expression of hepatitis B virus vector encoding TC-tagged core protein

摘要: Virus tagged with green fluorescent protein (GFP) contributes to the visualization and study of the virus in living cells. However, the hepatitis B virus (HBV) particle, which is a compact virion with limited internal space, cannot be incorporated with GFP tag as a large fragment. It was recently reported that protein genetically inserted with a smaller size tetracysteine (TC) tag could be specially labeled by a biarsenical fluorescent dye in living cells. In this study, we constructed a recombinant HBV vector encoding TC-tagged core protein for biarsenical labeling of HBV virion. TC tag was genetically inserted near the immunodominant c/e1 site of HBV core protein by mutagenesis. Western blot and enzyme-linked immunosorbent assay (ELISA) analysis showed that the TC-tagged core protein, hepatitis B surface antigen (HBsAg) and hepatitis B e antigen (HBeAg) could be expressed in cells transfected with the recombinant HBV vector, which is similar to the cells transfected with wild-type HBV vector. Reverse transcription-polymerase chain reaction (RT-PCR) and Southern blot analysis showed that HBV virion formation was affected by the genetic insertion of TC tag into core protein in some degree, but cells transfected with the HBV vector could still produce HBV virions incorporated with TC-tagged core proteins. Taken together, the recombinant HBV vector can serve as a useful tool to produce HBV virions incorporated with TC-tagged core proteins to be fluorescently labeled by biarsencial dye for visualizing and studying HBV in living cells.

Systems understanding of plant–pathogen interactions through genome-wide proteinprotein interaction

摘要: Plants are frequently affected by pathogen infections. To effectively defend against such infections, two major modes of innate immunity have evolved in plants; pathogen-associated molecular pattern-triggered immunity and effector-triggered immunity. Although the molecular components as well as the corresponding pathways involved in these two processes have been identified, many aspects of the molecular mechanisms of the plant immune system remain elusive. Recently, the rapid development of omics techniques (e.g., genomics, proteomics and transcriptomics) has provided a great opportunity to explore plant–pathogen interactions from a systems perspective and studies on protein–protein interactions (PPIs) between plants and pathogens have been carried out and characterized at the network level. In this review, we introduce experimental and computational identification methods of PPIs, popular PPI network analysis approaches, and existing bioinformatics resources/tools related to PPIs. Then, we focus on reviewing the progress in genome-wide PPI networks related to plant–pathogen interactions, including pathogen-centric PPI networks, plant-centric PPI networks and interspecies PPI networks between plants and pathogens. We anticipate genome-wide PPI network analysis will provide a clearer understanding of plant–pathogen interactions and will offer some new opportunities for crop protection and improvement.

Postprocessor development for ultrasonic cutting of honeycomb core curved surface with a straight blade

摘要: When ultrasonically cutting honeycomb core curved parts, the tool face of the straight blade must be along the curved surface’s tangent direction at all times to ensure high-quality machining of the curved surface. However, given that the straight blade is a nonstandard tool, the existing computer-aided manufacturing technology cannot directly realize the above action requirement. To solve this problem, this paper proposed an algorithm for extracting a straight blade real-time tool face vector from a 5-axis milling automatically programmed tool location file, which can realize the tool location point and tool axis vector conversion from the flat end mill to the straight blade. At the same time, for the multi-solution problem of the rotation axis, the dependent axis rotation minimization algorithm was introduced, and the spindle rotation algorithm was proposed for the tool edge orientation problem when the straight blade is used to machine the curved part. Finally, on the basis of the MATLAB platform, the dependent axis rotation minimization algorithm and spindle rotation algorithm were integrated and compiled, and the straight blade ultrasonic cutting honeycomb core postprocessor was then developed. The model of the machine tool and the definition of the straight blade were conducted in the VERICUT simulation software, and the simulation machining of the equivalent entity of the honeycomb core can then be realized. The correctness of the numerical control program generated by the postprocessor was verified by machining and accuracy testing of the two designed features. Observation and analysis of the simulation and experiment indicate that the tool pose is the same under each working condition, and the workpieces obtained by machining also meet the corresponding accuracy requirements. Therefore, the postprocessor developed in this paper can be well adapted to the honeycomb core ultrasonic cutting machine tool and realize high-quality and high-efficient machining of honeycomb core composites.

Establishment and verification of a shrinking core model for dilute acid hydrolysis of lignocellulose

摘要: The kinetics of lignocellulose hydrolysis under the conditions of high temperature and dilute acid (mass fraction 0.05%) was investigated in this paper. By studying the reducing sugar concentration versus reaction temperature (170°C–220°C) and reaction time (150–1800 s) during the hydrolysis process of five kinds of crop straw (rice, wheat, cotton, rape and corn), the shrinking core model was established, and the differential equation of the model and its analytical solution were obtained. With a numerical calculation method, the kinetic equation was estimated, and the degradation of reducing sugar obeyed first-order kinetics was obtained. The calculated results from the equations agreed well with the original experimental data. The calculation by the model showed that the reducing sugar concentration increases as the size of the particles decrease, and the uniform particles increase.

摘要: Heterogeneous catalysis with core-shell structures has been a large area of focus for many years. This paper reviews the most recent work and research in core-shell catalysts utilizing noble metals, specifically gold, as the core within a metal oxide shell. The advantage of the core-shell structure lies in its capacity to retain catalytic activity under thermal and mechanical stress, which is a pivotal consideration when synthesizing any catalyst. This framework is particularly useful for gold nanoparticles in protecting them from sintering so that they retain their size, structure, and most importantly their catalytic efficiency. The different methods of synthesizing such a structure have been compiled into three categories: seed-mediated growth, post selective oxidation treatment, and one-pot chemical synthesis. The selective oxidation of carbon monoxide and reduction of nitrogen containing compounds, such as nitrophenol and nitrostyrene, have been studied over the past few years to evaluate the functionality and stability of the core-shell catalysts. Different factors that could influence the catalyst’s performance are the size, structure, choice of metal oxide shell and noble metal core and thereby the interfacial synergy and lattice mismatch between the core and shell. In addition, the morphology of the shell also plays a critical role, including its porosity, density, and thickness. This review covers the synthesis and characterization of gold-metal oxide core-shell structures, as well as how they are utilized as catalysts for carbon monoxide (CO) oxidation and selective reduction of nitrogen-containing compounds.

The role of protein kinase C epsilon in neural signal transduction and neurogenic diseases

Protein kinase C epsilon (PKC ?) is one of major isoforms in novel PKC family. Although it has been extensively characterized in the past decade, the role of PKC ? in neuron is still not well understood. Advances in molecular biology have now removed significant barriers to the direct investigation of PKC ? functions in vivo, and PKC ? has been increasingly implicated in the neural biological functions and associated neurogenic diseases. Recent studies have provided important insights into the influence of PKC ? on cortical processing at both the single cell level and network level. These studies provide compelling evidence that PKC ? could regulate distinct aspects of neural signal transduction and suggest that the coordinated actions of a number of molecular signals contribute to the specification and differentiation of PKC ? signal pathway in the developing brain.

Wheat gluten protein and its impacts on wheat processing quality

Before the advent of the wheat genomic era, a wide range of studies were conducted to understand the chemistry and functions of the wheat storage proteins, which are the major determinants of wheat flour the suitability of wheat flour for various end products, such as bread, noodles and cakes. Wheat grain protein is divided into gluten and non-gluten fractions and the wheat processing quality mainly depends on the gluten fractions. Gluten provides the unique extensibility and elasticity of dough that are essential for various wheat end products. Disulfide bonds are formed between cysteine residues, which is the chemical bases for the physical properties of dough. Based on the SDS-extractability, grain protein is divided into SDS-unextractable polymeric protein (UPP) and SDS-extractable polymeric protein. The percentage of UPP is positively related to the formation of disulfide bonds in the dough matrix. In the wheat genomic era, new glutenins with long repetitive central domains that contain a high number of consensus hexapeptide and nonapeptide motifs as well as high content of cysteine and glutamine residues should be targeted.

Experiment and calculation on seismic behavior of RC composite core walls with concealed steel truss

摘要: To improve the seismic performance of reinforced concrete core walls, reinforced concrete composite core walls with concealed steel truss were proposed and systemically investigated. Two 1/6 scale core wall specimens, including a normal reinforced concrete core wall and a reinforced concrete composite core wall with concealed steel truss, were designed. The experimental study on seismic performance under cyclic loading was carried out. The load-carrying capacity, stiffness, ductility, hysteretic behavior and energy dissipation of the core walls were discussed. The test results showed that the seismic performance of core walls is improved greatly by the concealed steel truss. The calculated results were found to agree well with the actual measured ones.

Effects of inclination angles of disc cutter on machining quality of Nomex honeycomb core in ultrasonic

摘要: Ultrasonic cutting with a disc cutter is an advanced machining method for the high-quality processing of Nomex honeycomb core. The machining quality is influenced by ultrasonic cutting parameters, as well as tool orientations, which are determined by the multi-axis machining requirements and the angle control of the cutting system. However, in existing research, the effect of the disc cutter orientation on the machining quality has not been studied in depth, and practical guidance for the use of disc cutters is lacking. In this work, the inclined ultrasonic cutting process with a disc cutter was analyzed, and cutting experiments with different inclination angles were conducted. The theoretical residual height models of the honeycomb core, as a result of the lead and tilt angles, were established and verified with the results obtained by a linear laser displacement sensor. Research shows that the residual height of the honeycomb core, as a result of the tilt angle, is much larger than that as a result of the lead angle. Furthermore, the tearing of the cell wall on the machined surface was observed, and the effects of the ultrasonic vibration, lead angle, and tilt angle on the tear rate and tear length of the cell wall were studied. Experimental results revealed that ultrasonic vibration can effectively decrease the tearing of the cell wall and improve the machining quality. Changes in the tilt angle have less effect than changes in the lead angle on the tearing of the cell wall. The determination of inclination angles should consider the actual processing requirements for the residual height and the machining quality of the cell wall. This study investigates the influence of the inclination angles of a disc cutter on the machining quality of Nomex honeycomb core in ultrasonic cutting and provides guidelines for machining.

Construction of sustainable, colored and multifunctional protein silk fabric using biomass riboflavin

摘要: Riboflavin sodium phosphate has been confirmed as a promising biomass product derived from natural plants. In this paper, a novel method of dyeing and multifunctional modification of silk fabric by impregnation with riboflavin sodium phosphate was proposed, such that protein silk fabric can be endowed with bright yellow color and multi-functionality. The results of this paper confirmed that the pH and concentration of riboflavin sodium phosphate solution are critical factors for dyeing and multifunctional modification. Attractively, the photochromic performance was one of the most distinctive features of the modified silk fabric, and the dyed silk fabric turned into fluorescent green from original yellow under 365 nm ultraviolet lamp. Furthermore, the modified silk fabric exhibited good antibacterial properties with a high inhibition rate of 92% for Escherichia coli. Besides, the flame retardancy of silk fabric was significantly improved after modification. The damaged length of modified silk fabric with 40% owf riboflavin sodium phosphate was lower than 10.4 cm and passed the B1 classification. As revealed by the result of this paper, riboflavin sodium phosphate is sufficiently effective in serving as an eco-friendly multifunctional agent for strengthening the add-value of silk textiles.

Antithrombin deficiency and decreased protein C activity in a young man with venous thromboembolism:

Antithrombin and protein C are two crucial members in the anticoagulant system and play important roles in hemostasis. Mutations in and lead to deficiency or dysfunction of the two proteins, which could result in venous thromboembolism (VTE). Here, we report a Chinese 22-year-old young man who developed recurrent and serious VTE in cerebral veins, visceral veins, and deep veins of the lower extremity. Laboratory tests and direct sequencing of and were conducted for the patient and his family members. Coagulation tests revealed that the patient presented type I antithrombin deficiency combined with decreased protein C activity resulting from a small insertion mutation c.848_849insGATGT in and a short deletion variant c.572_574delAGA in . This combination of the two mutations was absent in 400 healthy subjects each from southern and northern China. Then, we summarized all the mutations of the and gene reported in the Chinese Han population. This study demonstrates that the combination of antithrombin deficiency and decreased protein C activity can result in severe VTE and that the coexistence of different genetic factors may increase the risk of VTE.

Improving performance portability for GPU-specific OpenCL kernels onmulti-core/many-coreCPUs by analysis-based

摘要: OpenCL is an open heterogeneous programming framework. Although OpenCL programs are functionally portable, they do not provide performance portability, so code transformation often plays an irreplaceable role. When adapting GPU-specific OpenCL kernels to run on multi-core/many-core CPUs, coarsening the thread granularity is necessary and thus has been extensively used. However, locality concerns exposed in GPU-specific OpenCL code are usually inherited without analysis, which may give side-effects on the CPU performance. Typically, the use of OpenCL’s local memory on multi-core/many-core CPUs may lead to an opposite performance effect, because local-memory arrays no longer match well with the hardware and the associated synchronizations are costly. To solve this dilemma, we actively analyze the memory access patterns using array-access descriptors derived from GPU-specific kernels, which can thus be adapted for CPUs by (1) removing all the unwanted local-memory arrays together with the obsolete barrier statements and (2) optimizing the coalesced kernel code with vectorization and locality re-exploitation. Moreover, we have developed an automated tool chain that makes this transformation of GPU-specific OpenCL kernels into a CPU-friendly form, which is accompanied with a scheduler that forms a new OpenCL runtime. Experiments show that the automated transformation can improve OpenCL kernel performance on a multi-core CPU by an average factor of 3.24. Satisfactory performance improvements are also achieved on Intel’s many-integrated-core coprocessor. The resultant performance on both architectures is better than or comparable with the corresponding OpenMP performance.
